Therapeutic Advances in Neurological Disorders

چکیده
Levetiracetam (LEV) is a new antiepileptic drug that is clinically effective in generalized and partial epilepsy syndromes as sole or add-on medication. Nevertheless, its underlying mechanism of action is poorly understood. It has a unique preclinical profile; unlike other antiepileptic drugs (AEDs), it modulates seizure-activity in animal models of chronic epilepsy with no effect in most animal models of acute seizures. Yet it is effective in acute in-vitro ‘seizure’ models. A possible explanation for these dichotomous findings is that LEV has different mechanisms of actions, whether given acutely or chronically and in ‘epileptic’ and control tissue. Here we review the general mechanism of action of AEDs, give an updated and critical overview about the experimental findings of LEV’s cellular targets (in particular the synaptic vesicular protein SV2A) and ask whether LEV represents a

Ionotropic Glutamate Receptors and their Role in Neurological Diseases
Glutamate is extensively and relatively uniformly distributed in the central nervous system (CNS) and its effects mediated by two distinct groups of receptors including Ionotropic and metabotropic glutamate receptors. Concentration of glutamate in the nervous system is much higher than in other tissues. Glutamate receptors play an important role in synaptic transmission, neural plasticity and n...
متن کاملChallenges in developing new multiple sclerosis therapies.
In the last 15 years, we have seen the therapeutic landscape for multiple sclerosis (MS) change dramatically. From a disease that essentially had no disease-modifying therapy, we have seen the approval of several drugs that have reduced relapses and altered the progression of disability in patients with MS. However, despite these advances, there is still no cure for the disease and the need for...
متن کاملTreatment of Neurological and Psychiatric Disorders with Deep Brain Stimulation Raising Hopes and Future Challenges
The technology of Neural Stimulation in recent years has become the focus of the research and treatment, although it has been around for many years. The potential use of stimulating the brain and nerves ranges from the spinal cord stimulation to the implantations of cochlear and bionic eyes with a large discrepancy between the clinical readiness for these various uses. Electrical high-frequency...
متن کاملP118: Metformin; A Review to its Anti-Neuroinflammatory Properties
Neuroinflammatory disorders include a wide spectrum of disorders in which immune system injures components of the nervous system. Despite the advances in therapy, lack of efficient curative therapies for these disorders not only affects the quality of life but also places huge burden on the society. These limitations have necessitated therapeutic interventions for developing more efficient regi...
متن کاملMetabotropic glutamate receptors and their ligands applications in neurological and psychiatric disorders
Metabotropic glutamate receptors (mGluRs) consist of a large family of G-protein coupled receptors that are critical for regulating normal neuronal function in the central nervous system. The wide distribution and diverse physiological roles of various mGluR subtypes make them highly attractive targets for the treatment of a number of neurological and psychiatric disorders. The discovery of ...
متن کاملSymptomatic therapy in multiple sclerosis.
Multiple sclerosis is the most common disabling neurological disease of young adults. The ability to impact the quality of life of patients with multiple sclerosis should not only incorporate therapies that are disease modifying, but should also include a course of action for the global multidisciplinary management focused on quality of life and functional capabilities.
